T h e s e data support the notion that pyrimidine dimers interrupt normal D N A replication and that synthesis r e s u m e s b e y o n d the dimer. Buhl et al. (1974) s h o w e d that after UV-irradiation replication can eventually b y p a s s these blocking pyrimidine dimers. By 24 hr after UV-irradiation of mammalian cells, the D N A synthesized in these cells w a s in normal-sized s e g m e n t s , even though pyrimidine dimers w e r e still present. This evidence indicates that UV-induced thymine d i m e r s can block D N A replication and that this blockage can be o v e r c o m e eventually by postreplication repair p r o c e s s e s .
